Tools for Studying T-cell Activation and TCR Signaling
T-cell activation is a fundamental process of adaptive immunity and plays a central role in developing next-generation immunotherapies. Following antigen recognition by the T cell receptor (TCR), a cascade of intracellular signaling events drives naive T-cell activation, proliferation, and differentiation into effector and memory T cells. The magnitude and duration of signals can affect T-cell response outcomes. Fine-tuning T-cell activation to optimize adaptive immunity has emerged as a central objective in immunotherapy development, particularly in cancer.
Advances in immuno-oncology have led to the emergence of innovative therapeutic strategies, including bispecific T-cell engagers and adoptive cell therapies. Such approaches require robust and physiologically relevant tools to evaluate T-cell activation, antigen specificity, signaling pathways, and functional responses. Primary T cells are the most physiological cellular model, but the high variability in donors' genetics and steady-state immunity presents major robustness limitations. Consequently, standardized high-throughput platforms, such as engineered Jurkat reporter cell lines, have emerged as powerful tools for reproducible assessment of TCR specificity, avidity, and function.
