InvivoGen offers a collection of NF-κB/IRF dual reporter THP-1 cells for Toll-like receptor (TLR) studies

InvivoGen’s THP1-Dual™ TLR cells are human monocytic THP-1 cell lines engineered to provide a rapid, sensitive, and reliable platform for studying Toll-like receptor signaling and for screening TLR agonists and antagonists.

THP1-Dual™ TLR cells feature two inducible reporter genes for SEAP (secreted embryonic alkaline phosphatase) and Lucia luciferase. As a result, these cells allow the simultaneous study of TLR-dependent NF-κB and/or IRF activation, by assessing the activity of SEAP or the secreted Lucia luciferase.
THP1-Dual™ TLR cells either express naturally several TLRs (TLR2, TLR4, TLR5 and TLR8), overexpress a selection of TLRs (TLR3, TLR4, TLR7, TLR8, TLR9) or were knocked-out for some TLRs (TLR2, TLR4, TLR8). Upon TLR stimulation, NF-kB activity can be monitored using QUANTI-Blue™, a SEAP detection reagent, and IRF activity can be determined using QUANTI-Luc™ 4 Lucia/Gaussia, a Lucia luciferase detection reagent.

These cell lines are extensively tested for viability, stability, biological activity, and absence of mycoplasma to ensure strong and reproducible results. Moreover, we provide detailed handling and experimental procedures for all cell lines, to minimize the end-user's need for optimization or troubleshooting.
