Cy5 labeled poly(I:C) HMW
Poly(I:C) HMW Cy5 is a fluorescent labeled synthetic analog of double-stranded RNA (dsRNA). This pathogen associated molecular pattern - short PAMP - is associated with viral infection and a strong ligand for the Toll-like receptor 3 (TLR3).
Polyinosinic-polycytidylic acid high-molecular weight (poly(I:C) HMW) was chemically labeled by direct covalent coupling of a Cyanine 5 (Cy5) probe containing a reactive alkylating group onto to reactive heteroatom within the nucleic acids. This confers fluorescent properties to Poly(I:C) HMW whilst retaining TLR3 recognition properties (see figure).
Cyanine5 (Cy5) is a bright, far-red-fluorescent dye with excitation designed for use with the 633 nm or 647 nm laser lines. Its reduced photobleaching properties, low background fluorescence in biological samples, enhancing signal-to-noise ratio, and its compatibility with a wide range of detectors and imaging systems makes Cy5 a great tool to use in fluorescence microscopy, flow cytometry, and other fluorescence-based applications (see figures).
In order to facilitate the intracellular delivery, the ligand should be complexed with a cationic lipid transfection agent, such as LyoVec™.
