Coelenterazine-utilizing luciferase detection reagents

QUANTI-Luc™ 4 is a liquid ready-to-use bioluminescent assay reagent to measure the activity of Lucia® Luciferase and other coelenterazine-utilizing luciferases. In order to meet your practical needs, InvivoGen offers:

— QUANTI-Luc™ 4 Lucia/Gaussia: for the detection of secreted Lucia® and Gaussia luciferases
— QUANTI-Luc™ 4 Renilla: for the detection of intracellular Renilla luciferase

Lucia®, Gaussia, and Renilla luciferases catalyze the oxidation of their coelenterazine substrate with the emission of light (bioluminescence). The light signal produced correlates to the amount of luciferase protein expressed. It is quantified using a luminometer and expressed as relative light units (RLUs). 

Each lot is thoroughly tested using InvivoGen's Lucia® reporter cell lines. One tube of QUANTI-Luc™ 4 Reagent is sufficient for 5 x 96-well plates (standard procedure) or 2 x 1536-well plates (HTS procedure).

Our proprietary QUANTI-Luc™ 4 Reagent has been optimized to reduce the coelenterazine substrate auto-luminescence for a better detection range. The optimal pH value for maximal luciferase activity differs between Lucia®, Gaussia, and Renilla. The final pH of the reaction using our reagents confers one type of luminescence detection for Renilla and two types for Lucia/Gaussia (see figures):

► Flash luminescence emits light with high intensity for a few seconds. It is ideal for endpoint detection in small sample numbers, or when using microplate readers with reagent injectors.
► Glow luminescence emits light with stable intensity for over 10 minutes. It is ideal for high-throughput screening (HTS) (i.e., the typical duration to read a 1536-well plate is ~7 minutes) or when using a luminometer without injectors (end-point detection).

QUANTI-Luc™ 4 Lucia/Gaussia

• Flash detection: without Stabilizer. This option is ideal for monitoring the cellular response to weak agonists

• Convenient liquid formulation: allows extemporaneous dilution; no need for storing a working solution

QUANTI-Luc™ 4 Renilla

• Two-component kit: QUANTI-Luc™ 4 Reagent and QUANTI-Luc™ 4 Lysis Buffer
• Intracellular luciferase detection: assess the activity of Renilla intracellular luciferase after cell lysis
• Low background: allows a high signal-to-background ratio
• Convenient liquid formulation: allows extemporaneous dilution; no need for storing a working solution
