Biotin-labeled double-stranded B-DNA
Poly(dA:dT) Biotin is a biotin-labeled synthetic analog of B-DNA, known to activate various cytosolic DNA sensors (CDS). It was chemically labeled by covalent coupling of a biotin probe containing a reactive alkylating group. It can be used for pulldown assays, ligand binding studies, and pattern recognition receptor (PRR) activation (see figures).

Poly(dA:dT) is a poly(deoxyadenylic-deoxythymidylic) acid sodium salt and a repetitive double-stranded DNA (dsDNA) sequence of poly(dA-dT)•poly(dT-dA). It is a synthetic analog of B-DNA, the most stable structure of DNA under ordinary cellular conditions. Intracellular poly(dA:dT) is detected by several CDS, such as cGAS, triggering the production of type I interferons (IFNs). This induction of IFN appears to be mediated by the endoplasmic reticulum protein STING. Moreover, poly(dA:dT) is recognized by AIM2 triggering the formation of an inflammasome and the subsequent secretion of pro-inflammatory cytokines, including IL-1β and IL-18.
To achieve stimulation of cytosolic PRRs, Poly(dA:dT) Biotin must be delivered into the cytoplasm, for example by using a transfection agent, such as LyoVec™.
