TRIF (TICAM1) is an adapter involved in the MyD88-independent signaling pathway of several TLRs. A dominant-negative form of TRIF was shown to completely abolish TLR3-induced signaling and also to block the NF-kB activity mediated by TLR2, TLR4 and TLR7 [1]. Further analysis has revealed that TRIF binds preferentially to TLR3 and TLR4 and is essential in their signaling pathways [2]. It predominantly activates IRF-3 and the production of IFN-b rather than NF-kB [3].

TRIF-DN

Dominant negative TRIF (aa387-566) harbors only the TIR domain and lacks the N-terminal and C-terminal portions [1]. Furthermore, it contains a proline to histidine mutation at amino acid 434. This proline residue is conserved among TLRs and is essential for activation of the TLR-mediated signaling.
