TRIF-related adaptor molecule (TRAM), also known as TIR-containing adaptor molecule (TICAM)-2, is a recently characterized adaptor molecule involved in the MyD88-independent TLR4 signaling pathway [1]. In response to LPS stimulation, TRAM physically bridges TLR4 and TRIF transmitting LPS-induced signaling to TRIF which in turn activates IRF-3 and the production of interferon beta [2]. TRAM function is restricted to the TLR4 pathway, as TRAM-deficient mice are defective in cytokine production in response to TLR4 ligand but not to other TLR ligands [3].
TRAM-DN
Dominant negative TRAM/TICAM2 (C117H) was generated by mutating the cysteine at position 117 into a histidine. This TRAM-DN was shown to retain its ability to bind to TLR4 but to lose its homo (TICAM2-TICAM2) and hetero (TICAM1-TICAM2) dimerizing properties [2].
