Interleukin-1 receptor-associated kinase (IRAK)-M is an inactive kinase found mainly in cells of the monocytic lineage , whereas other IRAKs are ubiquitous. IRAK-M negatively regulates TLR signaling. TLR stimulation induces IRAK-M which in turn down-regulates TLR signaling by preventing the dissociation of IRAK-1 and IRAK-4 from MyD88 and the formation of IRAK-1/TRAF6 complexes . IRAK-M-deficient cells exhibit increased cytokine production upon LPS stimulation and reduced endotoxin tolerance. IRAK-M is also involved in peptidoglycan-induced tolerance .
