Engineered Mouse Reporter RAW 264.7 Macrophages

InvivoGen offers RAW reporter cells derived from RAW 264.7, a mouse macrophagic cell line. It was originally established from a tumor in a BALB/c mouse induced with the Abelson leukemia virus. This adherent cell line is a commonly used model of mouse macrophages for the study of cellular responses to microbes and their products. RAW 264.7-derived cells endogenously express many PRRs, including toll-like receptors (TLRs), NOD-like receptors (NLRs), RIG-I-like receptors (RLRs), and C-type lectin receptors (CLRs). Of note, RAW 264.7 cells are naturally deficient for the expression of the canonical inflammasome adaptor protein ASC (apoptosis-associated speck-like protein containing a CARD domain).

All our cell lines are extensively tested for viability, stability, biological activity, and absence of mycoplasma to ensure strong and reproducible results. Moreover, we provide detailed handling and experimental procedures for all cell lines, to minimize the need for optimization or troubleshooting by the end-user.

InvivoGen's RAW-Blue™, RAW-Lucia-ISG™, and RAW-Dual™ reporter cells feature the inducible reporter genes for SEAP (secreted embryonic alkaline phosphatase), Lucia luciferase, or both, respectively. As a result, these cells allow the study of the NF-κB and/or the IRF pathways, by assessing the activity of SEAP or the secreted Lucia luciferase. Both reporter proteins are readily measurable in the cell culture supernatant when using QUANTI-Blue™, a SEAP detection reagent, and QUANTI-Luc™ 4 Lucia/Gaussia, a Lucia luciferase detection reagent.

The RAW inflammasome test cells feature a stable expression of the ASC adaptor protein, otherwise absent in RAW 264.7 cells. To help you assess the role of specific inflammasome actors in the canonical and non-canonical responses, cell lines deficient for the expression of gasdermin D (GSDMD), caspase 11 (CASP11), and NLRC4 were derived from the RAW-ASC cell line. In these cells, the NF-κB-inducible ASC::GFP reporter gene allows the monitoring of ASC-dependent inflammasome activation by visualizing GFP specks using fluorescence microscopy.

A collection of RAW knockout cell lines has been generated in the RAW-Lucia™ ISG cell line for genes involved in the signaling of TLR3, TLR4, or nucleic acids sensors.

Our RAW autophagy reporter cell line was designed to monitor the autophagic flux using a fluorescent-labeled fusion protein.
