Fc-Fusion Proteins
pFUSE-Fc is a family of plasmids developed to facilitate the construction of Fc-Fusion proteins by fusing a sequence encoding a given protein to the Fc region of an immunoglobulin. This region comprises the CH2 and CH3 domains of the IgG heavy chain and the hinge region. The hinge serves as a flexible spacer between the two parts of the Fc-Fusion protein, allowing each part of the molecule to function independently.
Fc-Fusion proteins can be readily purified by single-step protein A or protein G affinity chromatography (except mIgG1 which has low protein A binding affinity and may not work, depending on the fused protein).

Choose a pFUSE-Fc plasmid accordingly to your application:
• Protein purification - Most pFUSE-Fc can be used for Protein A or Protein G affinity chromatography.
• Long term expression in vivo - Choose a pFUSE-Fc with an Fc engineered to display an increased half-life
• Therapeutic use with cell depletion activity - Choose a pFUSE-Fc with an Fc engineered to display increased ADCC and CDC
• Therapeutic use without cell depletion activity - Choose a pFUSE-Fc with an Fc engineered to display reduced ADCC and CDC

Native Fc Regions
Warning: If you are considering using protein A for purification of mouse IgG1 Fc-fusion proteins, you may have to optimize purification condition as affinity is weak. High salt concentration and alkaline pH are expected to increase affinity [1]. Alternatively you can use protein G, that has a stronger affinity for mouse IgG1 but your fraction must be free from high affinity antibodies such as bovine IgG (fetal calf serum).
