Dual Expression Cassette Plasmids for the Expression of One Gene of Interest
pSELECT plasmids offer all the features necessary to express a gene of interest at high levels in a large number of cell types. pSELECT plasmids contain two transcription units, the first drives the expression of the gene of interest and the second drives the expression of a large choice of dominant selectable markers for both E. coli and mammalian cells.

Description
Two Convergent Expression Cassettes pSELECT contains two expression cassettes that are oriented in convergent directions. They are both terminating with a strong polyadenylation signal (polyA) that separates the two expression cassettes thus preventing any transcriptional interference. The late SV40 polyA terminates the transcription of your gene of interest while the human β-globin polyA terminates the transcription of the selectable marker.
Strong and Ubiquitous Expression of the Transgene pSELECT plasmids feature the strong EF1α/HTLV composite promoter that combines the elongation factor 1 alpha core promoter and the 5‘ untranslated region of the Human T-cell Leukemia Virus. This composite promoter confers high levels of expression in a constitutive manner in a wide range of mammalian cells.
Many Selectable Markers Available pSELECT plasmids come with a variety of selectable markers that confer antibiotic resistance in both E. coli and mammalian cells, as they are driven by the CMV promoter in tandem with the bacterial EM7 promoter.

Multiple Cloning Site or Reporter Gene pSELECT plasmids contain a multiple cloning site (MCS) downstream of the EF1α/HTLV composite promoter for convenient cloning of a gene of interest. All restriction sites of the MCS are compatible with other restriction sites, increasing the cloning options. pSELECT plasmids are also available with the LacZ reporter gene and can be used as control vectors. The size of pSELECT plasmids varies from 3165 bp to 6909 bp.
