TIR-deleted Forms of the TLR Genes
TLRs activate intracellular signaling pathways that share much in common with IL1-receptor (IL1-R) signaling, owing to their conserved Toll/IL1-R (TIR) domains present in the cytoplasmic tails. The TIR family also includes adapters, such as MyD88 and TRIF, that are recruited to the TLRs through their TIR domains, eliciting a signaling cascade leading to the activation of NF-κB. InvivoGen has engineered TIR-deleted TLR genes (TLR-∆TIR) that act as dominant negatives to serve as a useful tool to study TLR signaling.
Description
Expression of TLR-∆TIR Genes
A fragment of approximately 500 bp located at the 3‘ end of each TLR gene has been deleted to generate TLR-∆TIR genes that lack the TIR domain. These truncated genes are still able to recognize their ligands but are unable to induce the signaling cascade.
Easy Subcloning of the TLR-∆TIR Genes
To facilitate the excision and subcloning of a TLR-∆TIR gene from a pZERO-TLR plasmid to another vector, each gene is flanked by unique restriction sites that are compatible with many others.
Cotransfectable with pUNO or pDUO Plasmids
Each pZERO-TLR can be used for transient or stable transfection experiments. pZERO-TLR plasmids are selectable with puromycin in both E. coli and mammalian cells. Therefore, they can be cotransfected with a pUNO or pDUO plasmid.

Each pZERO-TLR plasmid is provided as a lyophilized transformed E. coli strain on a paper disk. Transformed strains are shipped at room temperature and should be stored at -20°C. Lyophilized E. coli cells are stable for at least 1 year when properly stored.
