Description

• Native promoters, also called minimal promoters, consist of a single fragment from the 5’ region of a given gene. Each of them comprises a core promoter and its natural 5’UTR. In some cases, the 5’UTR contains an intron.
• Composite promoters combine promoter elements of different origins (eg: SV40 enhancer/AFP promoter) or were generated by assembling a distal enhancer with a minimal promoter of the same origin (eg: CEA enhancer/promoter).

Two Backbones with Different Reporter Genes
In each pDRIVE, the promoter drives the expression of a reporter gene to facilitate the evaluation of its activity. Two different reporter genes are available in two different backbones:
• LacZ in pDRIVE: pDRIVE plasmids contain the LacZ reporter gene which expression can be determined using chromogenic, luminescent or histochemical detection.
• SEAP in pDRIVE5-SEAP: pDRIVE5-SEAP plasmids feature an engineered secreted embryonic alkaline phosphatase (SEAP) reporter gene. SEAP expression levels can be assayed with chromogenic or luminescent methods.

pDRIVE plasmids are selectable with the antibiotic Zeocin™ in E. coli. They can be used for transient transfection experiments in mammalian cells in vitro or for expression studies in vivo.

Contents and Storage

Each pDRIVE plasmid is provided as lyophilized transformed E. coli strain. Transformed bacteria are shipped at room temperature. Store at -20°C. Transformed bacteria are stable for at least 1 year when properly stored.
Each pDRIVE is provided with 4 pouches of E. coli Fast-Media® Zeo (2 TB and 2 Agar).
